Serval substrates have been identified, including cyclin-dependent kinase inhibitors (such as p21, p27, and p57), A20 and PARP1. It is a binding target for small molecules (like nimbolide) with potentials in targeted protein degradation drug development.

A. 3 ug 6xHIS-RNF114 separated in a 4-12% Bis-Tris gel (Coomassie-stained).
B. 6xHIS-RNF114 catalyzed polyubiquitin chain synthesis. The reaction containe 50 nM UbE1, 2 µM UbE2D4, 1 µM 6xHIS-RNF114, and 50 µM ubiquitin. Proteins wer separated in a 4-12% Bis-Tris gel and Coomassie-stained. Polyubiquitin chains (marked with asterisks) were synthesized. RNF114 autoubiquitination was not evident in Coomassie staining.
