USP30(64-502)

USP30(64-502) contains the catalytic domain of USP30 with additional loops being deleted. This variant is stable and highly active compared to the commonly used USP30(57-517) (ref. 4). USP30 deubiquitinates substrates of Parkin including MIRO1, TOMM20, MFN1, and MFN2 on mitochondrial membranes and functions to suppress mitophagy. It prefers cleavage of K6- and K11-linked polyubiquitin chains.

Coomassie-stained SDS-PAGE
(A) Lane 1: Molecular weight markers
Lane 2: 5 µg purified USP30(64-502)
(B) Reactions contained 10 nM USP30(64-502) + 0.5 uM Ub-Rhodamine 110 or Ub-Rhodamine 110 alone in DUB buffer II (cat# H1001). Rhodamine110 fluorescence was measured using a Synergy HTX plate reader with the excitation and emission filter set at 485/20 nm and 528/20 nm, respectively.
