Cy5 is covalently conjugated on lysines of recombinant protein G. The molar ratio between fluorescein and protein G in different lots is ~1 - 2. Protein G has higher affinity than protein A in binding of mouse, human, rat, goat and cow IgG.

Cy5-protein G recognizes various antibodies and can serve as the acceptor in TR-FRET assays that offer a sensitive tool in monitoring molecular proximity, post-translational modifications, enzyme activity and protein abundance. The donor is usually a europium-based chelator.

Cy5-protein G can also be used in varous fluorescent assays, including fluorescent western blotting and immunofluorescence microscopy.
