Details
This product contains an authentic K48-linked isopeptide bond, synthesized by using an enzymatic reaction containing UbE1, E2-25K, Ub(K48R) and Ub-fluorescein. Fluorescein is labeled on the C-terminus of Ub by mutating glycine 76 to a cysteine. In fluorescein labeled K48 Ub2, fluorescein is labeled on the proximal Ub. It is an excellent tool for monitoring deubiquitination that can be visualized by in-gel fluorescent detection of the production of fluorescein labeled monoUb (excitation at ~490 nm, and see product data sheet for examples). The assay is more quantitative and sensitive than immunoblotting of Ub. The detection limit is 10 ng or less.
