Pomalidomide is a second generation immunomodulator, and a thalidomide analog. Pomalidomide is a ligand of the E3 ubiquitin ligase Cereblon (CRBN), and it is widely used in PROTAC design for targeting protein substrates to the ubiqitin ligase CUL4/CRBN for ubiquitination, leading to subsequent proteasomal degradation.
