b-AP15

b-AP15 is a specific inhibitor of the deubiquitinating enzymes Uch37 and Usp14 of the 26S proteasome. It blocks the deubiquitinating activity of the 26S proteasome. Treating cells with b-AP15 causes the accumulation of polyubiquitinated proteins, inhibition of proteasomal degradation, upregulation of apoptotic markers and cell death.

Equal amount of whole cell lysates prepared from DMSO (lane 1) or 2 μM b-AP15 (lane 2)-treated HeLa cells were separated by SDS-PAGE and immunoblotted with an anti-Ub antibody. HEK293TCells were treated with DMSO or b-AP15 for 5 hours.
